اثرات سیستم‌های گابائرژیک و مورفینرژیک بر هستهCA1 هیپوکامپ روی رفتارهای اضطراب و حافظه برانگیخته بر اساس الگوی تست- تست مجدد

چکیده
هیپوکامپ، ساختاری کلیدی در پردازش اضطراب و ذخیره حافظه است. هدف از پژوهش حاضر، بررسی برهمکنش میان سیستم‌های گابائرژیک ناحیهCA1 هیپوکامپ و سیستم مورفینرژیک در تعدیل این رفتارها است. در این مطالعه موش‌های نر نژاد ویستار با استفاده از جراحی استرئوتاکسیک در ناحیه CA1کانول گذاری شدند. پس از طی دوره ریکاوری مورفین (آگونیست گیرنده‌های اوپیوئیدی مو) به صورت درون صفاقی، موسیمول و بیکوکولین (آگونیست و آنتاگونیست گیرنده‌هایGABAA) به درون CA1 تزریق گردید. پارامترهای مدت زمان حضور (%OAT) و تعداد ورود به بازوی باز (%OAE) در دستگاه EPM محاسبه شدند. آزمون تحلیل واریانس یک طرفه نشان داد تزریق درون صفاقی مورفین (%OAT) و (%OAE) را افزایش می‌دهد که حاکی از اثرات اضطراب زدای مورفین است. طبق نتایج آزمون t زوجی تشکیل حافظه بهبود یافت. تزریق پیش از آزمون دوز‌های مختلف موسیمول و بیکوکولین به ترتیب رفتارهای شبه اضطراب زدا و اضطراب زایی را القا نمود در حالیکه اثرات آنها روی حافظه بهبودبخش بود. تزریق سیستمیک دوز موثر مورفین به همراه دوزهای سه گانه موسیمول اثرات هم افزایی را در تشکیل حافظه و کاهش اضطراب القا نمود. تزریق این دوز از مورفین به همراه دوزهای سه گانه بیکوکولین، سبب تغییر اثرات منفی بیکوکولین روی اضطراب گردید ضمن اینکه تشکیل حافظه در این گروه‌ها افزایش یافت. این یافته‌ها نشان می‌دهند که سیستم‌های گابائرژیک و اپیوئیدرژیک تاثیرات مشابهی برحافظه و اضطراب در ناحیه CA1 دارند.

Effects of gabaergic and morphinergic systems in CA1 nucleus on anxiety and arousal memory based on test-retest paradigm

Hippocampus is a key structure in anxiety processing and memory storage. The purpose of this study was to investigate the interaction between GABAergic systems in CA1 area of hippocampus with morphinergic system in modulating of these behaviors. In this study, male wistar rats were cannulated with stereotaxic surgery in CA1 site. After a recovery period, morphine (µ opioid receptor agonist) was injected intraperitoneal, muscimol and bicuculline (GABAA receptor agonist and antagonist) were injected into the CA1. Parameters such as percentage of Open arms time (%OAT) and entries (%OAE) were calculated by means of an EPM apparatus. One-way ANOVA analysis showed that intraperitoneal administration of morphine increased %OAT and %OAE, indicating the anxiolytic effect of morphine. Based on paired sample T-test, the rate of memory formation improved. Pre-test intra-CA1 infusion of muscimol and bicuculline induced anxiolytic-like and anxiogenic-like behaviors, respectively; meanwhile, both drugs improved memory. Systemic injections of an effective dose of morphine combined with triple doses of muscimol showed a synergistic effect on memory formation and anxiety reduction. Simultaneous injection of morphine and bicuculline altered the negative effects of the latter on anxiety and increased the memory formation. This finding showed that GABAergic and opioidergic systems have similar effects on memory and anxiety in CA1 area.
