نقش کنترل منفی ژن شبه استریکتوسیدین سینتاز-7 در مقاومت گیاه آرابیدوپسیس تالیانا به تنش شوری

چکیده
ژن­های شبه­استریکتوسیدین سینتاز یک خانواده ژنی آرابیدوپسیس است که ژن­های اورتولوگ آنها در بقیۀ گیاهان کدکنندۀ آنزیم کلیدی مسیر بیوسنتزی ایندول آلکالوئیدهای مونوترپن هستند. بیان ژن SSL7 آرابیدوپسیس بلافاصله پس از تیمار با مولکول­ های پیام­ رسانی نظیر SA، متیل جازمونات و اتیلن القا می­ شود. در این آزمایش به­ منظور مطالعۀ کارکرد ژن شبه­ استریکتوسیدین سینتاز - 7 (SSL7) از ژنوتیپ وحشی گیاه آرابیدوپسیس تالیاناCol-0 (شاهد) و لاین سرکوب ­شدۀ ژن SSL7 (ssl7) از طریق درج T-DNA استفاده شد. تحلیل داده ­ها برمبنای آزمایش فاکتوریل در قالب طرح کاملاً تصادفی انجام شد. فاکتورها شامل دو ژنوتیپ و پنج سطح شوری شامل صفر، 50، 100، 150 و 200 میلی ­مولار NaCl بود. سرکوب ژن SSL7 در جهش ­یافته ssl7 با استفاده از qRT-PCR تأیید شد. بیان ژن­ های واکنش­ گر به تنش شوری شامل P5CS1، NCED3، AAO3 و RD29A در زمان­های 3 و 6 ساعت پس از اعمال تیمار NaCl در غلظتmM 150 نشان داد که چهار ژن مذکور در موتانت ssl7 در هر دو زمانِ پس از تیمار میزان بیان بالاتری نسبت به مقدار بیان در ژنوتیپ Col-0 نشان­ دادند. میزان فعالیت تعدادی از آنزیم ­های آنتی­اکسیدان در غلظت­ های مختلف NaCl در هر دو ژنوتیپ اندازه­ گیری شد. میزان فعالیت آنزیم‌های پراکسیداز (POD)، آسکوربات پراکسیداز (APX) و سوپر اکسید دیسموتاز (SOD) در موتانت ssl7 به ­طور معنی­ داری در سطح احتمال 1 درصد بیشتر از Col-0 بوده است. برعکس، فعالیت آنزیم‌ کاتالاز (CAT) در موتانت کاهش یافت. بنابراین، نتایج نشان داد که ژن SSL7 احتمالاً در مسیر مربوط به پاسخ به تنش شوری وابسته به سالیسیلیک ­اسید در گیاه آرابیدوپسیس نقش دارد و می­تواند به طریق کنترل منفی عمل کند.

چکیده English

Strictosidine synthase-like (SSL) is a group of gene families in the Arabidopsis genome, which whose orthologues in other plants are key enzymes in mono-terpenoid indole-alkaloid biosynthesis pathway. The SSL7 is upregulated upon treatments of Arabidopsis plants with signaling molecules such as SA, methyl jasmonate and ethylene. To find the functional role of the gene, a T-DNA-mediated knockout mutant (ssl7) along with the wildt ype were treated with different concentrations of NaCl. The expression level of salt stress genes including P5CS1, NCED3, AAO3 and RD29A at 150 mM NaCl demonstrated that the expression was significantly higher in ssl7 compared with the expression in Col-0. The activities of Catalase (CAT), Ascorbate Peroxidase (APX), Peroxidase (POD) and Superoxide Dismutase (SOD) were measured in different concentrations of NaCl. The results suggested that the enzymes activities were significantly higher in ssl7 compared with wild-type Col-0. In total, the results suggest that SSL7 might have a salicylic acid-dependent negative regulatory role in plant resistance to salt stress.
