میانکنش سیستم هیستامینرژیک هیپوکامپ پشتی و اپیوئیدرژیک سپتوم میانی بر رفتارهای شبه اضطرابی

چکیده
مهم­ ترین بخش مغز که در اضطراب نقش دارد، سیستم لیمبیک است که واجد سه بخش مهم هیپوکامپ، آمیگدال و سپتوم است. سیستم سپتوهیپوکامپ، نقش مهمی در تنظیم رفتارهای ترس و اضطراب برعهده دارد. بررسی­ ها نشان می­دهد که سیستم اپیوئیدرژیک نیز در رفتارهای شبه اضطرابی دخیل است. در این مطالعه اثر تزریق هیستامین به هیپوکامپ پشتی و تزریق مواد اپیوئیدرژیک به سپتوم میانی بر رفتارهای شبه اضطرابی در موش ­ها با استفاده از آزمونElvated Plus Maze (EPM) تحت بررسی قرار گرفت. تزریق µg/rat 5 و 1 هیستامین در هیپوکامپ پشتی اثری بر رفتارهای شبه اضطرابی نداشت، درحالی­که تزریق µg/rat10 هیستامین باعث افزایش درصد زمان ورود و دفعات ورود به بازوی باز شد، که نشان­ دهندۀ اثر اضطراب­ زدایی هیستامین است. تزریق مورفین (آگونیست رسپتور µ-اپیوئیدی) به درون سپتوم میانی (µg/rat1) باعث افزایش زمان ورود و دفعات ورود به بازوی باز شد. تزریق مقادیر µg/rat 25/ . و 5/ . مورفین اثری بر اضطراب نداشت. تزریق هم­زمان مقادیر بی­اثر هیستامین (µg/rat1) به هیپوکامپ پشتی و مورفین (µg/rat5/.) به سپتوم میانی باعث افزایش زمان ورود و دفعات ورود به بازوی باز شد. تزریق مقادیر مختلف نالوکسان (µg/rat 4و1،2) به­ عنوان آنتاگونیست گیرنده ­های اپیوئیدی به سپتوم میانی در حضور و غیبت مقدار مؤثر هیستامین (µg/rat10) در هیپوکامپ پشتی، تحت بررسی قرار گرفت. تزریق مقدار µg/rat4 نالوکسان به سپتوم میانی درصد زمان گذرانده در بازوی باز و درصد ورود به بازوی باز را کاهش داد، که این یافته­ ها نشان­ دهندۀ اثر اضطراب­ زایی نالوکسان بعد از تزریق به سپتوم میانی بود. تزریق مقدار µg/rat 10 هیستامین به هیپوکامپ پشتی و به­ طور همزمان تزریق سه مقدار مختلف نالوکسان (µg/rat4و2و1) نشان داد هیستامین اثر اضطراب­ زایی نالوکسان را با افزایش زمان ورود و تعداد ورود به بازوی باز، تا حدود زیادی خنثی کرد، هرچند در مقدار µg/rat4 نالوکسان این اثر کمتر است. از این یافته­ ها می­ توان نتیجه گرفت که سیستم هیستامینرژیک هیپوکامپ و اپیوئیدرژیک سپتوم میانی درسیستم سپتوهیپوکامپ با هم برهم­کنش دارند و برهم­کنش این دو سیستم در تعدیل اضطراب دخیل است.

Interaction between dorso-hyppocampal histaminergic and medio-septal opioidergic systems in anxiety behavior

چکیده English

Septohippocampal system plays an important role in regulating fear and anxiety behaviors. In this study, the effects of histamine injected into the dorsal hippocampus and opioidergic agents into medial septum on the anxiety-like behaviors in rats were analyzed, using the Elevated Plus-Maze (EPM) test. Injection of 1 and 5 μg/rat histamine into dorsal hippocampus had no effect on anxiety-like behavior, while injection of 10 μg/rat histamine increased the percentage of open arm time (%OAT) and open arm entry (%OAE), which indicated the anxiolytic effects of histamine. Microinjection of morphine, μ-opioid receptor agonist, into the medial septum (1μg/rat) increased the (%OAT) and (%OAE). Doses of 0.25, 0.5 μg/rat morphine had no effect on anxiety. Co-administration of histamine ineffective dose (1μg/rat) to the dorsal hippocampus and ineffective dose of morphine (0.25µg/rat) to the medial septum increased the (%OAT) and (%OAE). Subsequently, injection of different doses of naloxone (1, 2, 4 µg/rat), as an opioid receptor antagonist, into the medial septum in the presence and absence of an effective dose of histamine (10 µg/rat) in the dorsal hippocampus, was studied. Injection of naloxone (4 µg/rat) into medial septum decreased the (%OAT) and (%OAE), but did not alter the locomotor activity, which indicated the anxiogenic effects of naloxone. Simultaneous injection of histamine (10 µg/rat) into dorsal hippocampus with doses of naloxone (1, 2, 4 µg/rat) into the medial septum, indicate anxiolytic effects and increased %OAT and %OAE in Elevated Plus Maze, although when the dose of naloxone was 4μg/rat, this effect was less observed. The results indicate that hippocampus histaminergic system interact with medial septum opioidergic system and the interaction of these systems modulates anxiety behavior.
