برهم کنش میکوریز وزیکولار آرباسکولار با فلز سنگین نیکل و تأثیر آن بر تنش اکسیداتیو در گیاه جو

چکیده
حضور فلزهای سنگین باعث کاهش رشد گیاهان و نقص عمل­کرد آنها می ­شود. فلزهای سنگین همچنین، موجب القای تشکیل انواع فعال اکسیژن می­شوند و ازاین راه به سلول­ های گیاهی آسیب می­رساند. یکی از راهکارهای اساسی گیاه برای مقابله با آسیب­های ناشی از فلزهای سنگین، راه ­اندازی سیستم پاداکساینده­ است که خود شامل آنتی­ اکسیدان­ های آنزیمی و غیرآنزیمی است. از مهم­ ترین این نوع آنزیم­ ها، سوپراکسید­دیسموتاز است که با تبدیل آنیون سوپراکسید به پراکسید هیدروژن به بقای گیاه کمک می­کند. در ادامه، آنزیم ­های پراکسیداز و کاتالاز با تبدیل پراکسید هیدروژن به آب آن را خنثی می­کند. راهکار دیگر گیاه برای مقابله با فلزهای سنگین، هم­زیستی با قارچ میکوریزی وزیکولار آرباسکولار است. در پژوهش حاضر، بذرهای جو رقم ریحان در دو دستۀ میکوریزی و غیرمیکوریزی در چهار غلظت صفر، 100، 200، 300 و 400 میکروگرم گرم سولفات نیکل آب­دار بر گرم خاک در گلخانه کاشته شدند. پس ­از نمونه­برداری از بخش­های هوایی گیاهان مشخص شد در تیمارهای میکوریزی فعالیت آنزیم­های آنتی­اکسیدان در مقایسه با گیاهان غیرمیکوریزی بیشتر است. همچنین مشخص شد در ریشۀ گیاهان میکوریزی تجمع فلز نیکل در مقایسه با ریشۀ گیاهان غیرمیکوریزی بیشتر است که این خود نشان­ دهندۀ نقش قارچ میکوریز وزیکولار آرباسکولار در ممانعت از انتقال فلزهای سنگین و جلوگیری از آسیب به بخش ­های هوایی گیاه است.

چکیده English

Heavy metals reduce the plant growth and adversely affect plant performance potential. Heavy metals also induce the formation of reactive oxygen species leading to cell damage. Plants deal with heavy metal stress by activating enzymatic and non-enzymatic antioxidant systems such as Superoxide Dismutase which converts superoxide anion to hydrogen peroxide. Then Peroxidase and Catalase convert hydrogen peroxide to water. Another method employed by pl-ants to deal with heavy metals is mycorrhizal symbiosis. In this study, mycorrhizal and non-mycorrhizal barley seedlings (Hordeum vulgar), var. Rayhan, grown in a greenhouse were treated with four concentrations of nickel (0,100,200 and 400 µg/g). Results showed greater antioxidant activity in mycorrhizal plants compared with non-mycorrhizal plants. Also, it was determined that the accumulation of nickel in roots of mycorrhizal plants was greater compared with non-mycorrhizal plants.
