بررسی روند تکامل صفات ریخت شناسی در گونه های ایرانی سردۀ Stachys از نعنائیان براساس توالی هسته ای nrITS

چکیده
تکامل صفات ریخت ­شناختی در چارچوب مطالعات تبارزایشی و براساس داده­ های توالی nrITS صورت گرفت. در این تحلیل 28 آرایه شامل دو برون گروه (Betonica officinalis, Melittis mellisophyllum) و 26 آرایه از گونه ­های سرده Stachys که در ایران رویش دارند به­ منزلۀ درون­گروه انتخاب شدند. به­منظور بررسی روند تکامل صفات، ده صفت رویشی و زایشی انتخاب شد. این صفات شامل زیستگاه، شکل رویشی، ریخت‌شناسی برگ، گل‌آذین، کاسه، و جام کرک­پوش بودند. بازسازی روابط تبارزایشی داده­ های توالی nrITS براساس روش استنباط بیزی انجام شد. سپس، بررسی روند تکامل صفات ریخت­ شناختی با استفاده از نرم ­افزار Mesquite و ماتریس داده­ های ریخت­ شناسی انجام گرفت. نوع صفت، حالت­ های صفت، تعداد گام­ های و ضریب لازم و ضریب سازگاری برای هریک از صفات محاسبه شد. از میان صفات ریخت­ شناختی بررسی ­شده، صفات مربوط به شکل رویشی و وجود کرک­پوش ساده از جمله صفاتی بودند که دارای بالاترین ضریب سازگاری بود و کمترین جورگرایی را نشان می­دادند. کمترین میزان ضریب سازگاری به صفاتی همچون کاسۀ متقارن، برگ­های قاعده­ ای پایا و وجود کرک غده ­ای اختصاص داشت و به عبارت دیگر بیشترین میزان جورگرایی را نشان می­دادند. بررسی روند تکامل صفات ریخت­ شناختی نشان داده است که بخشۀ Eriostomum و بخشۀ Fragilicaulis به ­ترتیب از جمله ابتدایی ­ترین و پیشرفته ­ترین بخشه­ های سرده Stachys به شمار می رود.

Investigation of the evolutionary trend of morphological characters of Stachys (Lamiaceae) in Iran based on nrITS sequences data

The evolutionary trend of the morphological characters of the genus Stachys in Iran, on the basis of nrITS se-quence data, was investigated. A total of 28 nrITS sequences, representing 26 species of Stachys as well as Betonica of-ficinalis and Melittis mellisophyllum, were obtained from GenBank. Patterns of character evolution were assessed for 10 vegetative and reproductive characters with emphasis on taxonomic treatments of Stachys performed earlier to infer its history and interpret processes of change. The most important characters included habitat, growth form, leaf morp-hology, inflorescence, calyx and corolla as well as trichome morphology. Phylogenetic tree, based on the nrITS dataset, were constructed by Bayesian analysis. Parsimony mapping was performed in Mesquite v. 1.12. The present analysis revealed that the state of symmetrical calyx, the presence of basal leaves as well as the presence of glandular trichomes were not in agreement with the results of molecular data, which indicated the artificial nature of these characters in pr-evious classification. The presence of simple trichomes as well as the growth forms were consistent with phylogeny ba-sed on nrITS sequences. Evolutionary trend of morphological characters demonstrated that the sect. Eriostomum and sect. Fragilicaulis were the most primitive and the most advanced sections in Iran, respectively.
