بهینه سازی تولید آنزیم لیپاز در Salinivibrio sp. SA2 با طراحی تاگوچی

چکیده
آنزیم لیپاز یکی از مهم­ ترین آنزیم­ های هیدرولیتیک است که در صنایع مختلف مانند صنایع غذایی، ‌لبنیات، ‌دارویی،‌ شوینده­ ها و غیره کاربرد دارد. در این آزمایش، به­ منظور بهینه ­سازی تولید لیپاز در باکتری Salinivibrio sp. SA2، از روش طراحی آزمایش تاگوچی به­منزلۀ روشی قدرتمند برای بررسی متغیرهای مؤثر‏ بر فرآیند تولید آنزیم، میان­کنش فاکتورهای مختلف و به­ دست ­آوردن ترکیبی بهینه از متغیرهای تحت آزمایش استفاده شد. شرایط بهینه برای فاکتورهای pH، ‌دما، دور شیکر، غلظت روغن زیتون و نوع نمک به­ترتیب 8،°C 35، rpm 100، 2 درصد و کلرید سدیم M 1 به­ دست آمد. عوامل با تأثیر معنی­ دار بر فرآیند تولید لیپاز به­ ترتیب اهمیت pH، هوادهی و نمک تعیین شدند. حداکثر فعالیت لیپاز در شرایط بهینه و در سطح معنی­ داری 5 درصد (p< 0.05U/mg 4/120 محاسبه شد.

Lipase is one of the most important hydrolytic enzymes widely used in various commercial activities such as food, dairy, pharmaceutical and detergent inducteries. In this experiment, Taguchi method was attempted as a powerful method to optimize the factors affecting enzyme production and to investigate the interactions among these factors and their optimum combination in Salinivibrio sp. SA2. The optimum conditions for pH, temperature, shaker's rpm, olive oil concentration and salt type turned out to be 8, 35 °C, 100 rpm, 2% and sodium chloride 1 M, respectively. Significant factors influencing on the lipase production proved to be pH, agitation and Salt type. The maximum lipase activity in optimum condition and at the 5% significance level (p< 0.05) was 120.4 U/mg.
