fa بررسی اثرات هم افزایی کورکومین با داروی سیس‌پلاتین بر بیان ژن Cdc42 در سلول‌های رده Calu-6 کارسینومای ریه در انسان Study of Cisplatin effects in comparison with Curcumin on Cdc42 gene expression in human Calu-6 lung carcinoma cell line علوم سلولی و مولکولی Cell and Molecular Biology مقاله پژوهشی Original Article  سرطان ریه دومین سرطان شایع است. ناکارآمد بودن درمانها و عوارض جانبی داروهای شیمی درمانی، اهمیت مسیر درمان را صدچندان مینماید. هدف این مطالعه بررسی اثر کورکومین و مقایسه اثر آن با داروی سیسپلاتین و نیز اثر ترکیب همزمان آنها بر رده سلولی Calu-6 و بیان ژن Cdc42 است. رده سلولی اپیتلیالی Calu-6 رده سلولی کارسینومای ریه انسان است که قبل و بعد از تیمار با دوز  µg/ml 0/5 تا 8 کورکومین و  µg/ml 0/1 تا 50 سیس‌پلاتین و تیمار با ترکیب هر دو طی 24 و 48 ساعت، شمارش و میزان بقای سلولی به‌وسیله روش MTT بررسی شد. برای بررسی آپوپتوز و تغییر بیان ژن Cdc42، که یک GTPase از خانواده Rho ها است و در سرطانهای مختلف افزایش مییابد، از تکنیکهای فلوسایتومتری و Real time PCR استفاده و دادهها توسط نرمافزارهای GraphPad prism، Flowing software، Linreg، Excel و Rest آنالیز شد. در سلولهای تیمار شده با دوز µg/ml 0/67 کورکومین و <a name="OLE_LINK1">µg/ml</a> 1/7 سیس‌پلاتین (کمترین دوزهای موثر) طی 24 ساعت کاهش زنده مانی سلولها و همچنین القای مرگ سلولی نیز مشاهده شد به‌طوریکه بیشترین مقدار آپوپتوز اولیه و آپوپتوز با تاخیر به ترتیب مربوط به تیمار با کورکومین و ترکیب همزمان کورکومین و سیسپلاتین بود. همچنین هم سیسپلاتین و هم کورکومین باعث بروز تغییرات مورفولوژیکی در سلولها شدند. نتایج آنالیز ژن نشان داد کورکومین و سیسپلاتین باعث کاهش بیان ژن Cdc42 میشوند. در تمامی موارد اثرات سینرژیک کورکومین و سیسپلاتین مشاهده شد. بنابراین کورکومین می تواند گزینه خوبی برای استفاده ترکیبی با داروهای شیمیدرمانی به منظور کاهش دوز مصرفی دارو و کاهش عوارض جانبی آنها باشد.   <gdiv></gdiv><gdiv></gdiv><gdiv></gdiv><gdiv></gdiv> Lung carcinoma is the second most common type of cancer. Inefficiency of the current treatments and the undesirable side effects of chemotherapy drugs made the know-how of the treatment important. The purpose of this study is to investigate the synergic effect of curcumin and Cisplatin in comparison with the sole application of each treatment on Calu-6 cell line, an epithelial cell line of human lung carcinoma, and the expression of Cdc42 gene. The viability of Calu-6 was examined after 24- or 48-hour treatment with doses of 0.5 to 8 µg/ml of curcumin, 0.1 to 50 µg/ml of cisplatin and combined doses of curcumin and Cisplatin by MTT assay. To measure apoptosis and the expression of Cdc42 gene, flow cytometry and Real-Time PCR were utilized. Decrease of cell viability and induction of cell death were observed in the cells treated with 0.67 µg/ml of curcumin and 1.7 µg/ml of cisplatin (the lowest effective dose) and the combined treatment with the same doses of each drug after 24-hour treatments. The maximum rates of early and late apoptosis were related to treatment with curcumin and the combined treatment. The gene expression analysis results indicated that both Curcumin and Cisplatin decrease the expression of Cdc42 gene, moreover, their co-administration showed synergic effects.
