اثر تنش های خشکی و شوری بر تولید هورمون IAA در باکتریهای جداسازی شده از خاک-های شور و شور-سدیمی
The effect of drought and salinity tensions on IAA production in isolated bacteria from saline and saline-sodic soils

<p dir="RTL" style="text-align: justify">این تحقیق به­ منظور ارزیابی توانایی تولید هورمون<span dir="LTR">Indole acetic acid (IAA) </span> به­ وسیلۀ باکتری­ های جداسازی شده از خاک­ های متأثر از خشکی و شوری اجرا شد. به­ همین منظور ضمن جداسازی 400 باکتری از خاک­ های با درجات مختلف قابلیت هدایت الکتریکی و نسبت سدیم تبادلی، تغییرات تولید <span dir="LTR">IAA</span>، اثر مقادیر مختلف تریپتوفان و تنش­ های خشکی و شوری بر تولید این هورمون تحت بررسی قرار گرفت. نتایج نشان داد که اثر تنش­ های خشکی و شوری بر تغییرات تولید <span dir="LTR">IAA</span> از نظر آماری معنی­ دار بود. گونۀ باکتری <em><span dir="LTR">Arthrobacter siccitolerans</span></em> (جداسازی شده از خاک­ های شور-سدیمی)، مولد هورمون <span dir="LTR">IAA</span> است و مقایسۀ میانگین داده­ ها نشان داد که این باکتری در وضعیت تنش خشکی به میزان 3/84 درصد نسبت به جدایۀ <span dir="LTR">RS11</span>، هورمون بیشتری تولید کرد. در­حالی­که جدایۀ <span dir="LTR">RS11</span> در شرایط بدون تنش خشکی در مرحله اول غربالگری، بیشترین مقدار <span dir="LTR">IAA</span> (<span dir="LTR"> µg.ml<sup>-1</sup></span>41/10) را نسبت به سایر باکتری­ها تولید کرد. همچنین <em><span dir="LTR">siccitolerans</span></em> <em><span dir="LTR">A</span></em><span dir="LTR">.</span> تا قابلیت الکتریکی برابر با 40 دسی<span dir="LTR">­</span>زیمنس بر متر و پتانسیل اسمزی 25- بار همچنان به تولید <span dir="LTR"> IAA</span> ادامه داد. مقدار تولید هورمون <span dir="LTR">IAA</span> به­ ازای مصرف هر واحد تریپتوفان، در این باکتری برابر با 0/1 و در باکتری­ های دیگر، کمتر از 0/1 بود. با توجه به آزمون<span dir="LTR">­</span>های انجام شده، برتری نسبی باکتری یاد­شده محرز شد و برای انجام آزمون<span dir="LTR">­</span>های مزرعه<span dir="LTR">­</span>ای در تنش­ های موجود در خاک­ های متأثر از خشکی، شوری و شور- سدیمی پیشنهاد شد.</p>
<p style="text-align: justify">This investigation was carried out to evaluate Indole acetic acid (IAA) production by isolated bacteria from drought and salinity affected soils. Four hundred bacteria were isolated from soils with different levels of electrical conductivity (EC) and sodium adsorption ratio (SAR). Then the ability of these bacteria to produce IAA and the effects of different amounts of tryptophan and drought-salinity tensions on IAA production were analyzed. Results indicated that the effects of drought and salinity tensions on variability of IAA production were statistically significant. One of the bacterial species isolated from saline-sodic soils, <em>Arthrobacter siccitolerans</em>, was IAA producer and data comparison indicated that under drought stress conditions it produced IAA 84.3% higher than isolate RS11. While isolated RS11 produced highest rate of IAA (10.42 µg.ml<sup>-1</sup>) under normal conditions. Also, <em>A.siccitolerans </em>continued to produce IAA until the EC was 40 dS.m<sup>-1</sup> and OP (Osmosis pressure) was -20 bar. The amount of IAA production per unit of applied tryptophan in this bacterium was 0.1 and in others it was lesser. According to experiments, the relative preference of this bacterium was verified and proposed for field experiments in drought, salinity and salinity-sodicity affected soils.</p>
