بررسی اثر ضد سرطانی کافئیک اسید در پوشش های کیتوزان و نانولوله کربنی بر رده سلول های سرطانی دهانه رحم رده HELA

چکیده
نانولوله های کربنی عامل دار شده می‌توانند به عنوان حامل‌های مولکولی در دارورسانی استفاده شوند. این ساختارها در درمان سرطان مورد استفاده قرارگرفته‌اند. مولکول‌های دارویی معمولاً به گروه‌های عاملی سـطحی نانولوله ‌ها یا پلیمرهای پوشش داده‌ شده بر روی نانولوله‌ ها متصل می‌شوند. هدف از این تحقیق، بررسی اثر نانولوله کربنی پوشش‌دار شده با کیتوزان حامل کافئیک‌ اسید بر سطح بیان ژن های Bax و Bcl-2، رشد و تکثیر سلولی در سلول های سرطانی دهانه رحم رده HELA است. از تست MTT، جهت بررسی میزان بقای سلولی استفاده شد. میزان بیان ژن Bax در نانولوله ‌های پوشش‌داده‌ شده با کیتوزان حاوی کافئیک اسید، نانولوله‌ های کربنی بدون پوشش با کافئیک اسید، نانولوله ‌های کربنی خام و کافئیک اسید به ترتیب 724/10، 696/6، 985/1 و 737/3 بود. میزان بیان ژن Bcl-2 در نانولوله ‌های پوشش‌داده‌ شده با کیتوزان حاوی کافئیک اسید، نانولوله ‌های کربنی بدون پوشش با کافئیک اسید، نانولوله ‌های کربنی خام و کافئیک اسید به ترتیب 138/0، 264/0، 749/1 و 399/0 بود. بر اساس نتایج به‌ دست ‌آمده، نانولوله کربنی پوشش‌دار شده با کیتوزان حامل کافئیک‌ اسید نسبت به نانولوله کربنی بدون پوشش کیتوزان، نانولوله ‌های کربنی و کافئیک‌ اسید به تنهایی باعث القای آپوپتوز، رشد و تکثیر سلولی در سلول‌های سرطانی HELA می‌گردد. هم چنین استفاده از کافئیک ‌اسید به عنوان دارو و نانولوله کربنی پوشش‌دار شده با کیتوزان حامل کافئیک ‌اسید می‌تواند به‌ عنوان استراتژی امیدوارکننده ای در درمان سرطان دهانه رحم مورد توجه قرار گیرد.

Investigation the anticancer effect of caffeic acid in carbon nanotube with and without chitosan coating on human cervical cancer HELA cell line

The aim of this study was to investigate the effect of cofactor-bearing chitosan-coated and non-chitosan carbon nanotubes on induction of apoptosis of cervical cancer HELAcell line by measuring the expression of Bax and Bcl-2 genes as well as cell growth and proliferation. MTT test was used for cell proliferation assay. Expression changes of Bax and Bcl-2 genes were analyzed by Real Time PCR. The MTT results showed that the carbon nanotube coated with caffeic acid carrier chitosan and without caffeic acid carrier and crude carbon nanotube and caffeic acid induce apoptosis and cell toxicity. The results showed that caffeic acid with a chitosan-coated carbon nanotube carrier increased Bax expression and decreased Bcl-2 gene expression, so that the expression ratio of Bcl-2 / Bax compared to free caffeic acid to significantly increase. The results show that the death of cancer cells treated with caffeic acid with chitosan-coated carbon nanotube carries an increase in Bcl-2 / Bax expression ratio and thus induces apoptosis in HELA cancer cells. . Also, the use of caffeic acid as a drug and carbon nanotube-coated carbon nanotube carriers of caffeic acid can be considered as a promising strategy in the treatment of cervical cancer.
