مقایسه تولید گاما- دکالاکتون در سویه طبیعی و جهش یافته مخمر Yarrowia lipolytica

چکیده
گاما-دکالاکتون استری حلقوی از اسیدهای چرب هیدروکسی است که دارای رایحه هلویی بوده و در صنایع غذایی و آرایشی کاربرد گسترده‌ای دارد. تولید زیست‌فناورانه این ترکیب با تبدیل زیستی روغن دانه کرچک توسط مخمر Yarrowia lipolytica امکان پذیر است. هدف تحقیق حاضر مقایسه تولید گاما-دکالاکتون توسط سویه طبیعی با سویه جهش یافته دارای قابلیت تولید لیپاز با مقادیر بالا است. بر اساس نتایج به دست آمده از این تحقیق، سلول‌هایی با مورفولوژی مخمری بیش از سلول‌های هیفی شکل گاما-دکالاکتون تولید می‌کنند. زمان اوج تولید نیز برای سویه طبیعی و جهش یافته به ترتیب 24 و 72 ساعت پس از تلقیح و بیشینه تولید گاما-دکالاکتون برای سویه طبیعی و جهش یافته به ترتیب 65 و 90 میلی گرم در لیتر به دست آمد. سویه جهش یافته 38 درصد سوبسترای بیشتری را نسبت به سویه طبیعی به گاما-دکالاکتون تبدیل می‌کند که بهره‌وری تولید در مقیاس صنعتی می‌تواند به صورت قابل توجهی افزایش یابد.

Gamma-decalactone, an intramolecular 4-hydroxydecanoic acid ester, has a peach-like aroma and is widely used in the food and cosmetics industries. The biotechnological production of this compound is possible via biotransformation of castor seed oil by the yeast Yarrowia lipolytica. This study aimed to compare the production of gamma-decalactone by wild-type strain with that in a mutant strain producing lipase in high amounts. It was found that cells with yeast-like morphology produce more gamma-decalactone than hyphae-like cells. The maximum production of gamma-decalactone by wild-type and mutant strains was 65 mg/L after 24h of inoculation and 90 mg/L after 72h of inoculation, respectively. The mutant strain converts 38% more substrate into gamma-decalactone than the wild-type strain, therefore, it could significantly increase the productivity of industrial-scale production of gamma-decalactone.
