تاثیر کوئرستین بر اختلال حافظه و استرس اکسیداتیو ناشی از مسمومیت با مالاتیون در موش­های صحرائی نر نژاد ویستار

چکیده
مالاتیون جزو رایج­ترین نوع حشره­کش ­های ارگانوفسفره است ­که استرس اکسیداتیو به عنوان یکی از مکانیسم­های ملکولی اصلی در سمّیت این ماده بر اندام­ های بدن شناخته شده است. کوئرستین یکی از ترکیبات فلاونوئیدی است. هدف مطالعه حاضر بررسی اثر آنتی­اکسیدانی کوئرستین بر حافظه فضایی و پارامترهای استرس اکسیداتیو موش­های نر نژاد ویستار طی مسمومیت با سم مالاتیون است. این مطالعه بر روی 9 گروه 8‎ تایی موش­های صحرایی نر صورت گرفته است. سه روز پس از تزریق درون صفاقی داروهای­ کوئرستین، مالاتیون و یا تداخل این داروها، دستگاه ماز آبی موریس به منظور سنجش پارامترهای مرتبط با حافظه فضایی استفاده شد. نمونه برداری از هیپوکامپ انجام گردید و پارامترهای استرس اکسیداتیو در این ناحیه سنجش گردید. تزریق درون صفاقی دوزهای mg/kg­ 200 و 100 مالاتیون به طور معنی­داری موجب کاهش پارامترهای حافظه فضایی (01/0 >P ) و نیز القا استرس اکسیداتیو شد (001/0 >P ). تزریق درون صفاقی دوز mg/kg 50 کوئرستین حافظه فضایی در رت­های مسموم شده با مالاتیون را بهبود بخشید (05/0 >P ). همچنین پارامترهای استرس اکسیداتیو درگروه­ های مسموم شده با مالاتیون، با تیمار کوئرستین کاهش معنی­داری یافت (01/0 > P). کوئرستین قادر به بازگرداندن عملکرد حافظه فضایی و مقدار پارامترهای استرس اکسیداتیو گروه­ های تحت تیمار با مالاتیون به سطح طبیعی است.

The effects of Quercetin on memory and oxidative stress impairment due to Malathion poisoning in male Wistar rats

Malathion is one of the commonest type of organophosphate insecticide whose toxicity in human body is mainly considered to result from the induction of oxidative stress. The aim of this study was to investigate the antioxidant effect of Quercetin, a flavonoid compound, on the spatial memory and oxidative stress parameters during Malathion poisoning in male Wistar rats. This study was performed on nine groups, each of which consisted of eight male rats. Three days after intra-peritoneal injection of Quercetin, Malathion or a combination of these two drugs, the Moris Water-Maze apparatus was used to measure spatial memory parameters. The hippocampus was sampled and the oxidative stress parameters were measured in this area. Intra-peritoneal injection of Malathion (100 and 200 mg/kg) significantly reduced spatial memory parameters (P<0.01) and induced oxidative stress (P<0.001), whereas intra-peritoneal injection of quercetin (50 mg/kg) improved spatial memory in Malathion-poisoned experimental rats (P<0.05). Also, oxidative stress parameters in Malathion-treated groups showed a significant reduction in quercetin treatment (P<0.01). Quercetin was observed to restore the function of spatial memory and the level of oxidative stress parameters of the treated groups with Malathion to the normal level.
