بررسی ارتباط ماهیان کور غار لرستان (Garra thyphlops و Garra lorestanensis) و جمعیت های ماهی گلچراغ (Garra gymnothorax) در حوضه های دز و کرخه

چکیده
غار محل زیست ماهیان کور لرستان در حوضه رود دز در مجاورت رودخانه سزار در منطقه ­ای با ساختار کارستی قرار گرفته است. در مجاورت رودخانه سزار در دیواره­ های دره رودخانه چشمه­ هایی وجود دارد که ممکن است امکان ورود ماهیان رودخانه ­ای را به داخل منابع آب زیرزمینی فراهم کنند. این مشاهدات احتمال رابطه مهاجرتی در بین ماهیان gymnothorax Garra رودخانه سزار و ماهیان کور typhlops Garra و Garra lorestanensis را پیش می ­آورد. علاوه بر این در بین ماهیان کور در مورد هر گونه، تنوع شکل بدن قابل مشاهده است. تنوع فنوتیپی یادشده ممکن است گویای تنوع ژنتیکی ناشناخته ­ای در بین این ماهیان باشد و یا ناشی از شرایط محیطی حاکم بر بخش­ های مختلف غار ماهی کور باشد. در این مطالعه برای پاسخ به سوالات موجود از روش­ های تعیین توالی ژن سیتوکروم اکسیداز زیرواحد یک (COI) و تعیین توالی نسل جدید استفاده شده است. نتایج نشان می­ دهد اشکال دوکی شکل و اشکال دارای بدن کشیده و غیر دوکی گونه ­های typhlops G. و lorestanensis G. از نظر ترکیب ژنوم و ژن میتوکندریایی تفاوتی ندارند. همچنین نتایج مقایسه ترکیب ژنومی ماهیان کور و ماهی gymnothorax G. رودخانه­ های دز و کرخه نشان می­ دهد احتمالا نفوذ ژنتیکی بسیار کمی (کمتر از 3 درصد) از گونه gymnothorax G. به ماهی کور typhlops G. وجود دارد. شاید بتوان این نفوذ ژنتیکی کم ماهیان سطح­ زی را به سازگاری پایین­ تر آن­ها برای ورود و انطباق با شرایط زیستگاه زیرزمینی نسبت داد.

چکیده English

The cave barb habitat is located in a Karst formation along the Sezar River. The springs on the walls of the Sezar River valley may provide a means for fish in surface waters to penetrate into the underground waters. These observations propose the probability for a migratory relationship between Garra gymnothorax in the Sezar River and the cave barbs (Garra typhlops and Garra lorestanensis). In addition, a variety of different body shapes including fusiform and slender body forms are observed among the cave fish. This phenotypical variation may be a sign of an unknown genetic diversity or could be attributed to the variable environmental conditions in different parts of the subterranean habitat. To clarify the situation, we used the sequences of mtDNA cytochrome oxidase subunit I and next generation sequencing method. The results showed that the fusiform and slender body shapes of G. typhlops and G. lorestanensis were not different with regard to their mtDNA and genomic compositions. Moreover, the analysis of the genomic their mtDNA and genomic compositions. Moreover, the analysis of the genomic showed that a limited level of gene flow (less than 3%) from G. gymnothorax probably existed in G. thyphlops. The low level of gene flow may be related to the lower fitness and adaptability of the surface dwelling fish to the subterranean life conditions.
