جداسازی باکتری های تولیدکنندۀ مواد آنتی باکتریال از خاک شور

چکیده
خاک ­های شور دارای پراکندگی وسیعی در کشور ایران هستند. این خاک­ های بکر منبع بسیار بارزشی برای جداسازی باکتری ­های جدید با متابولیت­های با کارکرد بالا در زیست فن­آوری هستند. سویه­ های اکتینومیست از محیط کشت­های نشاسته کازئین آگار و ISP2 با درصدهای مختلف از نمک سدیم کلرید (0، 5 و 10 درصد) از نمونه­ های خاک تیمارشده جدا شدند. کلنی ­های خالص باکتری ­ها، روی محیط کشت گلیسرول کازئین کشت داده شدند و پس از رشد کامل با لایه نازکی از مولر هینتون آگار (1 درصد) حاوی Staphylococcus aureus مقاوم به متی سیلین (MRSA) پوشیده شدند. متابولیت­ های فعال سویه ­های منتخب استخراج و به روش انتشار در ژل بررسی شدند. 38 درصد از جدایه­ ها تولیدکنندة ترکیبات متابولیتی با فعالیت آنتی ­بیوتیک علیه این پاتوژن بودند. متابولیت ­های تولیدشده توسط جدایه­ های act-2 و act-5 که دارای هاله عدم رشد موثرتری علیه MRSA بودند استخراج و اثر ضد MRSA عصاره act-5 نشان داده شد. همچنین اثر ضدمیکروبی سویه act-5 علیه سایر باکتری­ ها نیز بررسی شد و باکتری مورد نظر شناسایی شد. در این تحقیق برای اولین بار اکتینومیست­ های نمک دوست تولیدکننده ترکیبات فعال زیستی از خاک­ های شور شهرستان قم جداسازی شدند و پتانسیل تولید متابولیت­ های ضد MRSA توسط این میکروارگانیسم­ها تحت بررسی قرار گرفت. نتایج این تحقیق، پتانسیل اکتینومیست­ های خاک شور را برای تولید متابولیت­ های مفید نشان می­ دهد.

Isolation of antibacterial material-producing bacteria from saline soil

Saline soils are widely spread in Iran. These intact soils are a great source for the isolation of new bacteria with highly functional metabolites in biotechnology. Actinomycete strains were isolated on starch casein agar and ISP2 with different concentrations of sodium chloride (0, 5 and 10%) from treated soil samples. Pure colonies were cultured on a casein glycerol medium. After complete growth, the plates were covered with a thin layer of Muller Hinton Agar (1%) containing methicillin-resistant Staphylococcus aureus (MRSA). Active metabolites of selected strains were extracted and their antibacterial activities analyzed by agar well diffusion method. 38% of isolates produced antibiotics against the pathogen. The metabolites produced by act-2 and act-5 isolates, which had a more effective inhibition zone against MRSA, were extracted and anti MRSA activity of act-5 extract was shown. The antimicrobial activity of act-5 against other bacteria was also investigated and the bacterium was identified. In this study halophilic actinomycetes producing bioactive compounds were isolated from the saline soils of Qom and the anti-MRSA potential of their metabolites was investigated for the first time. The results of this study show the potential of saline soil actinomycetes for the production of useful metabolites.
