افزایش فعالیت آنزیم سلولاز در قارچ تریکودرما ویریده با القای جهش با پرتو گاما

چکیده
گونه ­های سرده Trichoderma ازنظرتولید سلولاز با فعالیت آنزیمی بالا بسیار شناخته شده هستند. با وجود تولید آنزیم‌های سلولازی متنوع در قارچ تریکودرما، به ­علت عمل­کرد آنزیمی پائین و هزینۀ زیاد تولید، استفاده از آنزی­م های سلولازی آنها در تبدیل زیستی پسماندهای سلولزی چندان موفقیت‌آمیز نبوده است. تحقیق حاضر با هدف دست­یابی به جدایه‌های موتانتی از گونۀ Trichodera viride با توان تولید بیشتر آنزیم سلولاز با استفاده از القای جهش با پرتو گاما انجام گرفته است. سوسپانسیون اسپور قارچ در اپتیمم دوز 250 گری تحت پرتوتابی با گاما قرار گرفت. اسپورهای جوانه ­زده به ­منظور بررسی پایداری پنج ­بار واکشت شدند و از بین آنها جدایه‌های موتانت با قابلیت اسپورزایی بهتر انتخاب شدند. فعالیت سلولازی با استفاده از سوبستراهای سلولزی کاغذ صافی واتمن شمارۀ یک، کربوکسی متیل سلولز، آویسل، سلولز باکتریایی و سلولز کلوئیدی و براساس دستورالعمل IUPAC سنجیده شد. پروفایل پروتئین‌های سلولازی جدایه‌های موتانت با آزمون SDS-PAGE بررسی شد. بیشترین فعالیت سلولاز کل و فعالیت آنزیم آویسلاز به ­ترتیب با 43/92 و 40/74 U/mg در جدایۀ موتانت M21 و بیشترین فعالیت اندوگلوکانازی در جدایۀ موتانت M18 مشاهده شد. به­ طور کلی کاربرد اشعۀ گاما در 38 درصد از جدایه­ های جهش‌یافته به افزایش معنی‌دار در فعالیت سلولازی منجر شد. همچنین، از این روش به ­منزلۀ روشی ساده و کارآمد برای دست­یابی به جدایه­ های موتانتی می­ توان استفاده کرد که­ توانایی تولید مقادیر بالای آنزیم و دیگر متابولیت ­های میکروبی را داشته باشند.

Trichoderma species have been famous for production of cellulases with relatively high enzymatic activity. However, attempts to use their cellulolytic enzymes in the bioconversion of cellulosic wastes have not been successful because of high cost of production and low enzymatic yields. This study aimed to obtain gamma-induced mutants of T. viride with enhanced extracellular cellulase production. Spore suspensions were exposed to γ-rays at 250 Gy as optimum dose. After irradiation, all germinated spores were grown onto PDA plates and mutant strains with better sporulation were selected and subcultured five times to test their stability. Cellulase activity was tested using Whatman No. 1 filter paper, carboxymethyl cellulose, avicel, bacterial cellulose and walseth cellulose according to the IUPAC recommendation. Extracellular proteins profiles of mutant strains were studied via SDS-PAGE. The maximum activity of total cellulase and avicelase were observed in the isolate of M21 (92.43 and 74.40 U/mg, respectively) and maximum endo-glucanase activity was observed in M18 mutant. The results of this study showed that the application of gamma ray led to a significant increase in Cellulose activity of 38 percent of mutant strains. Thus, this method could be used as a simple and efficient way to achieve strains with the ability to produce high levels of enzymes and other biological metabolites.
