ایجاد جهش نقطه ای در اسید آمینۀ 263 ژن استرپتوکیناز و کلونینگ و بیان پروتئین جهش یافتۀ حاوی سیستئین

چکیده
استرپتوکیناز یکی از شناخته ­شده ­ترین عوامل ترومبولیتیک با مصرف بالینی گسترده است. با وجود این، کاربرد آن به­ دلیل ایمونوژن بودن، ایجاد عوارض هموراژیک و نیمۀ­عمر نسبتاً کوتاه، همراه با خطرهایی است. پگیلاسیون اختصاصی بر روی اسیدآمینۀ سیستئین، تکنیکی مفید جهت کاهش بسیاری از این عوارض است. هدف از مطالعۀ حاضر، طراحی و تولید مولکول استرپتوکیناز جهش ­یافتۀ حاوی سیستئین است که برای پگیلاسیون اختصاصی قابل­ استفاده باشد. اسیدگلوتامیک 263 استرپتوکیناز، که یک اسیدآمینۀ سطحی در ساختار پروتئین استرپتوکیناز است، برای انجام جهش و تعویض با سیستئین انتخاب­شد. برای نیل به این هدف، با به­ کارگیری روش SOEing PCR، جهش بر روی کدون GLU263 ژن استرپتوکیناز برای تبدیل به کدون سیستئین انجام شد. سپس، ژن استرپتوکیناز دست ­نخورده و جهش­ یافته در وکتور بیانی pET-26b (+) وارد شدند. ساختارهای حاصل درون اشریشیا کلی Rosetta (DE3) ترنسفرم شده و توسط القا با IPTG بیان شدند. درنهایت، تولید پروتئین­ ها به­ وسیلۀ آزمون­ های SDS-PAGE و وسترن بلاتینگ تأیید شد. پروتئین­ ها به­ وسیلۀ افینیتی کروماتوگرافی با ستون Ni-NTA agarose در شرایط دناتوره با اوره تخلیص شدند و برای حذف اوره و تاخوردگی مجدد پروتئین­ ها از فیلتراسیون ژلی با سفادکس G-25استفاده شد. نتایج این مطالعه نشان داد که با استفاده از وکتور و میزبان مذکور ژن جهش­ یافتۀ حاوی کدون سیستئین به­ خوبی بیان می ­شود و برای پگیلاسیون اختصاصی مناسب خواهد بود.

چکیده English

Streptokinase is one of the best known thrombolytic agents with widespread clinical use. However, its use is not risk-free due to its immunogenicity, hemorrhagic complications and relatively short half-life in circulation. Specific PEGylation of cysteine residue is a useful technique for reducing most of these complications. The aim of this study was designing and producing a cysteine containing mutant of streptokinase, to be used for specific PEGylation. Glut-amic acid 263, which is a surface amino acid in the structure of streptokinase protein, was selected for replacement with cysteine amino acid by site directed mutagenesis. The Glu263 codon was changed to cysteine codon by SOEing PCR technique. Then, the intact and mutated streptokinase genes were inserted into expression vector pET-26b (+). The co-nstructs were transformed to Escherichia.coli Rosetta (DE3) strain and the proteins were expressed by IPTG induction. The proteins were confirmed by SDS-PAGE and western blot analysis, purified by Ni-NTA agarose affinity chroma-tography under denaturing condition with urea and Sephadex G-25 column was applied to remove urea to refold the pr-oteins. This study indicated that by using aforesaid vector and host, cysteine containing mutant gene is expressed well and it will be appropriate for specific PEGylation.
