تعیین حدود گونه‌ای کمپلکس .Linaria lineolata s.s در ایران بر پایه شواهد ریخت‌شناختی و مولکولی

چکیده جنس .Linaria Mill به‌عنوان یکی از گروه‌های پیچیده تاکسونومیکی در تیره Plantaginaceae، شامل گونه‌هایی است که تفکیک آن‌ها به‌دلیل شباهت‌های ریخت‌شناختی و تنوع درون‌گونه‌ای همواره با چالش همراه بوده است. در این مطالعه، چهار گونه از کمپلکس Linaria lineolata s.s. (شامل گونه‌های Linaria lineolata، Linaria elymaitica، Linaria karajensis و Linaria khalkhalensis) با استفاده از داده‌های ریخت‌شناسی و مولکولی مورد بررسی قرارگرفتند. در مجموع ۲۰ صفت کمی و کیفی در قالب تحلیل‌های آماری تک‌متغیره و تحلیل مؤلفه‌های اصلی (PCA) ارزیابی شد. به‌منظور مطالعه روابط مولکولی، توالی‌های ژن هسته‌ای کم‌نسخه AGT1 و ناحیه بین‌ژنی کلروپلاستی rpl32–trnL مورد استفاده قرار گرفت و روابط فیلوژنتیکی با روش استنتاج بیزین بازسازی شد. نتایج PCA نشان داد که گونه‌های مورد مطالعه در فضای رج‌بندی به‌طور مشخص از یکدیگر تفکیک می‌شوند و دو مؤلفه اول بخش عمده‌ای از واریانس داده‌ها را تبیین می‌کنند. همچنین تحلیل‌های فیلوژنتیکی مبتنی بر ژن هسته‌ای AGT1 و ناحیه بین ژنی کلروپلاستی rpl32–trnL نشان داد که تاکسون‌های مورد مطالعه در کلادهای مجزا قرار گرفته و الگوی تفکیک ژنتیکی با نتایج ریخت‌شناسی همخوانی دارد. بر پایه صفات ریخت‌شناسی دارای بیشترین ارزش تشخیصی، کلید شناسایی دوشاخه‌ای برای گونه‌های مورد مطالعه نیز تدوین شد. همخوانی میان داده‌های ریخت‌شناسی و مولکولی از استقلال تاکسونومیکی گونه‌های بررسی‌شده حمایت می‌کند. نتایج این مطالعه ضمن حمایت از حدود گونه‌ای اعضای کمپلکس L. lineolata s.s.، نشان داد که تلفیق شواهد ریخت‌شناختی و مولکولی می‌تواند رویکردی مؤثر برای روشن‌سازی روابط تاکسونومیکی و بازنگری کمپلکس‌های گونه‌ای در جنس Linaria باشد.

Molecular relationships in the Veronica hederifolia complex (Plantaginaceae, Veroniceae)

چکیده Veronica is a species-rich genus of the Plantaginaceae, comprising over 500 annual and perennial herbs distributed across both hemispheres. We examined relationships within the Veronica hederifolia complex (V. hederifolia, V. cymbalaria, V. panormitana, and V. triloba), all belonging to V. subg. Cochlidiosperma. Overlapping morphological features complicate taxonomic delimitation in this group. To clarify species boundaries, we analysed 28 sequences from three regions: the nuclear low-copy CYCLOIDEA2 region, the nuclear ribosomal internal transcribed spacer (ITS), and the plastid DNA (trnL–trnF), in addition to morphological characters. Morphological variation was observed in leaf shape, calyx and pedicel orientation, corolla diameter and colour, sepal form, and seed features. Bayesian phylogenetic inference revealed that the CYC2 marker provided the highest resolution, confirming the monophyly of the complex and distinguishing V. triloba as a separate species. V. cymbalaria and V. panormitana formed sister lineages, with V. panormitana as the earliest-diverging taxon. Plastid trnL–trnF sequences showed limited variability, and ITS data displayed partial incongruence, likely due to hybridization, introgression, and polyploidy. Overall, integrating molecular and morphological evidence highlights the utility of low-copy nuclear markers in resolving recently diverged and reticulate lineages, supporting distinct evolutionary trajectories within the V. hederifolia complex.
