از آنجایی که در سال ­های اخیر باکتری­ های اندوفیت گیاهی کاربردهای متنوع و مفیدی در بیوتکنولوژی پیدا کرده­اند، توجه زیادی به جداسازی، شناسایی و ارزیابی این نوع میکروارگانیسم ­ها شده است. در این مطالعه با توجه به دشوار بودن سترون سازی سطوح بافت­ های گیاهی از باکتری­ های اپی­فیت، کارایی سه روش مختلف غربالگری باکتری­های اندوفیت شامل 1- استریل کردن با آب ژاول، 2- استریل کردن متناوب (تندال تغییر یافته) و 3- استریل کردن با تریتون X100 و آب ژاول مورد ارزیابی قرار گرفت. روش تندال تغییر یافته روشی ابداعی است که در این پژوهش به منظور حذف مناسب اسپور­های درونی باکتری ­های اپی فیت که در جداسازی اندوفیت­ ها مانع به حساب می ­آیند، به کار رفت. بیش ­ترین تعداد باکتری­ های اندوفیت از گیاهان دولپه ­ای و از برگ جداسازی شدند. همچنین باکتری­ های اندوفیت از نظر تولید آنزیم ­های مختلف هیدرولازی بررسی شدند در این میان آنزیم پروتئاز در طیف وسیعی از باکتری­ های اندوفیت و به مقدار بیشتر از سایر آنزیم­ ها تولید شد. سویه EndoA، مورد شناسایی مولکولی قرار گرفت و مشخص شد که به میزان 100 درصد مشابه Bacillus halotolerans است.
