بررسی القای ریشۀ مویین در برخی از گونه های سردۀ مریم گلی

چکیده
القای ریشۀ مویین در گیاهان از طریق ورود T-DNA از پلاسمید Agrobacterium rhizogenes به ژنوم سلول­ های گیاهی انجام می­ شود. مطالعۀ حاضر با هدف بررسی اثر نوع سویۀ باکتری و گونۀ گیاهی در القای ریشۀ مویین در دو گونۀ بوم ­ویژۀ ایرانSalvia eremophila) و (Salvia reuterana و پنج گونۀ غیربوم ­ویژۀ Salvia macrosiphon)، Salvia multicaulis، Salvia nemorosa، Salvia verticellata و (Salvia virigata سردۀ مریم­گلی توسط چهار سویۀ باکتری شامل 4A، 15834ATTC، 1724، 2659 به صورت آزمایش فاکتوریل درقالب طرح کاملاً تصادفی انجام شد. ریزنمونه­ های ساقه و دم­برگ قادر به تولید ریشۀ مویین نبودند، درحالی­که تقریباً تمام ریزنمونه ­های برگی ریشۀ مویین تولید کردند. ماهیت تراریخت ریشه ­های مذکور از طریق ردیابی بخشی از ژن­ rol C واردشده به ژنوم گیاهی به اثبات رسید. نتایج نشان داد که سویه ­های مختلف A. rhizogenesو گونه­ های مختلف مریم­ گلی اثر معنی­داری بر تعداد و فراوانی ریشه­ های مویین دارند. بیشترین تعداد ریشۀ مویین به­ ازای هر ریزنمونه (12/5 ریشۀ مویین) و بالاترین فراوانی ریشه­ زایی (82%) در اثر تلقیح گونۀ S. macrosiphon با باکتری سویۀ 15834ATTC به­دست آمد. همچنین، بیشترین تعداد ریشۀ مویین به ازای هر ریزنمونه در گونه­ های S.eremophila (32/3 ریشۀ مویین) و S. reuterana (92/3 ریشۀ مویین) توسط باکتری4A، در گونه­ های S. nemorosa (6/2 ریشۀ مویین)، S. multicaulis (36/4 ریشۀ مویین) و verticillata S. ( 5 ریشۀ مویین) توسط سویۀ 2659 و در گونۀ­ S.virigata (3 ریشۀ مویین) توسط سویۀ 15834ATTC به­دست آمد. به­ طورکلی، به­ نظر می­رسد نوع گونۀ گیاهی، ریزنمونه و سویۀ باکتریایی مورد استفاده نقش به­ سزایی در میزان القای ریشۀ مویین در سردۀ مریم­ گلی دارد.

Investigation of hairy root induction in some Salvia L. species

چکیده English

Hairy root induction in plants is the result of the insertion of T-DNA from Agrobacterium rhizogenes into the plant genome. The present study was conducted to investigate the effect of bacterium strain and plant species type on hairy root induction in two endemic (Salvia eremophila and S. reuterana) and five non-endemic (S. macrosiphon, S. multicaulis, S. nemorosa, S. verticellata and S. virigata) Salvia by four bacteria strains including 1724, 2659, ATCC-15834 and A4. Petiole and stem explants were not capable of inducing hairy roots, while almost all leaf segments produced it. Confirmatory studies were carried out by direct detection of inserted rol C by the PCR. The results showed that different Agrobacterium rhizogenes strain and Salvia spices had significant effect on hairy roots number and frequency. The infection of S. macrosiphon via A. rhizogenes strain ATCC15834 showed the highest number of infe-cted roots per explant (5.12 hairy roots) and root frequency (82%). The highest number of hairy root per explant in S. eremophila (3.32 hairy roots) and S. reuterana (3.92 hairy roots) were achieved by inoculation with strain A4. Strain 2659 produced the highest hairy roots number in S. nemorosa (2.6 hairy roots), S. multicaulis (4.36 hairy roots) and S. verticillata (5 hairy roots). Also hairy roots formation occurred at the highest number in S. virigata (3 hairy roots) with infection by strain ATCC15834.
